Objectives
The purpose of this project is to develop a targeted long read sequencing based platform for the rapid detection of structural variants (SVs) in cancer genomes using Cas9-targeted technology.The project will involve the development of:(1)        A scalable bioinformatics platform for SV detection.(2)        Perform long read sequencing whole genome sequencing and higher-order chromatin interactions to identify functional SVs in acute lymphoblastic leukaemia (ALL).(3)        Develop PCR-free CRISPR/Cas9 based targeted sequencing panel for rapid SV detection.The methods involved will include bioinformatics analysis, nanopore long read sequencing and Cas9-based targeted excision of DNA for sequencing.This project will enable detection of genomic variants that cannot be accurately detected by existing methods. It will fill the gap in the field of cancer precision medicine, and especially benefiting ALL patients.
