Objectives
This project aims to generate the new type of human cytotoxic immune cells, ITNKs and CAR-ITNKs derived from human T cells and stem cells, to characterize them by performing in vitro assays and validating in preclinical models, and to explore T cell-derived CAR-ITNKs for the treatment of malignant tumors. The proposed preclinical and clinical research has four parts: 1. Development of low immunogenic human EPSCs for producing universal and ""off-the-shelf"" immune cells; 2. Generation of ITNKs and CAR-ITNKs from the genetically engineered human EPSCs; 3. Preclinical assessment CAR-ITNKs derived from T cells and from the engineered human EPSCs; 4. Clinical investigation with CAR-ITNKs derived from T cells, focusing on DLBCL therapy
