Abstract | In E. co the first step in biotin biosynthesis is the condensation of pimeloyl CoA with alanine to form 8amino-7-oxononanoic acid (AON). This reaction is catalysed by the pyridoxal phosphate-dependent enzyme AON synthase (AONS, bioF gene product). We have cloned, overexpressed and purified the enzyme and characterised the enzyme using a variety of spectroscopic techniques. We have now determined the structure of the enzyme to 1.9A using X-ray crystallography. We present details of the structure and mechanism of AONS, the first such analysis of an enzyme from the type II PLP family. |
