| Abstract | 利用隨機引物法反轉錄合成SRV-1 ss-cDNA和ds—cDNA.將ds—cDNA平端連接到質粒pUC19中,轉化E.Coli.DH5α菌.氨芐青霉素、X-gal培基和菌落雜交篩選,克隆陽性重組率為20.7%.EcoRⅠ、BamHⅠ雙酶切分析,12.7％的插入片段大于500bp。Southern雜交進一步確證,所選三株重組質粒(pSRV-127、pSRV-156和pSRV-169)的探針用插入片段(0.9kb、1.0kb和1.4kb),是SRV-1 cDNA片段.光敏生物素標記純化pSRV-169DNA和pSRV-156cDNA片段制備探針,前者用于檢測SRV-1病毒RNA,后者用于檢測純化病毒顆粒。堿性磷酸酶系統染色,靈敏度分別為18pg和4×105vp. |
