AbstractObjective: To investigate the effects of porphyromonas gingivalis-lipopolysaccharide (Pg-LPS) on the cell cycling of the human periodontal ligament cells ( hPDLCs) in vitro. Method: HPDLCs (3rd to 5th passages) were incubated with 0.01ng/ml Pg-LPS for 24 hours in vitro, and then the DNA synthesis in different phases of cell cycling of the hPDLCs were analyzed using the flow cytometry (FCM). Result: It was found that the G0/G1-phase fraction of hPDLCs was decreased significantly by Pg-LPS (p<0.01), while the proliferation index (PI) was increased (p<0.05 ). Both the S-phase fraction (SPF) and the G2/M-phase fraction were higher in hPDLCs incubated with Pg-LPS than those in control cells (p<0.05 for SPF; p<0.01 for G2/M-phase fraction). Conclusion: Pg- LPS with the assessed concentration enhances the proliferation of hPDLCs by augmenting the DNA synthesis in both S-phase and G2/M phase of cell cycling.
