AbstractExperimental autoimmune encephalomyelitis (EAE) has been successfully used as a model of multiple sclerosis. With the loss of myelin in CNS axons, the pathology involved generally affects the overall integrity of the white matter structure. In this study, radial and axial diffusivities was comprehensively determined by high-resolution DTI in the spinal cord (SC) samples of EAE induced rats, and compared to histological and behavioral analyses. Radial diffusivity was found to be sensitive to demyelination or myelin quantification. Relatively large variation of both radial and axial diffusivities was observed. In addition, SC degeneration was found to be spatially inhomogeneous. The results underscore the necessity of using volumetric DTI, and both radial and axial diffusivity analysis to accurately characterize the neurodegeneration in this disease.
