AbstractPIK3R1 (encoding the p85α subunit of phosphatidylinositol 3-kinase) is the 11th most frequently mutated gene across tumors. We recently reported neomorphic p85α mutants that induce signaling cascades not predicted by the canonical functions of p85α, suggesting the need to functionally annotate specific mutations in cancer genes for effective genome-informed personalized therapy.
